About Enclomiphene

Clomiphene is sold as a mix of two mirror-image forms, zuclomiphene and enclomiphene. Enclomiphene is the trans-isomer, the one that acts as a clean estrogen blocker at the hypothalamus and pituitary, while zuclomiphene behaves more like a weak estrogen and lingers much longer.

Repros Therapeutics developed the product citrate as Androxal for men with low testosterone of pituitary origin. The FDA didn’t approve it, so it remains a research compound.

Mechanism of action

Estrogen normally tells the hypothalamus and pituitary to ease off on GnRH, LH and FSH. Enclomiphene sits on those estrogen receptors without switching them on, so the brake comes off and the pituitary releases more LH and FSH.

In males, more LH drives the testes to make more testosterone, and more FSH supports sperm production. That’s the difference from giving testosterone directly, which shuts the same axis down.
