What is MOTS-c?

MOTS-c is a 16-amino-acid peptide coded not in the cell nucleus but in mitochondrial DNA, inside the gene for the 12S ribosomal RNA. Changhan Lee and colleagues in Pinchas Cohen’s lab at the University of Southern California described it in 2015.

It belongs to a small group of mitochondrial-derived peptides, alongside humanin, that act as signals from the mitochondria to the rest of the cell and body.

How MOTS-c works

Most proteins are coded in the cell nucleus. MOTS-c is one of a handful of peptides coded in mitochondrial DNA instead. Its main target looks like the folate cycle, which feeds into AMPK activation and better glucose handling in muscle.

Under stress it can move into the nucleus and change gene expression. Circulating levels drop with age in both mice and people, which is why it’s studied in metabolism and aging.

Research findings

The 2015 Cell Metabolism paper reported that MOTS-c prevented diet-induced obesity and insulin resistance in mice, working mainly through skeletal muscle.

Reynolds and colleagues reported in Nature Communications in 2021 that exercise raises MOTS-c levels in people, and that treating older mice with it improved their physical performance.

How MOTS-c compares

Short version: MOTS-c is a short peptide encoded in mitochondrial DNA that switches on AMPK. Labs often weigh it against these:

  • AOD-9604 is the 176-191 tail of growth hormone, studied for fat breakdown
  • GLP-2 TZ (GLP-2 TZ, tirzepatide) acts on two incretin receptors, GIP and GLP-1
  • GLP-3 RT (GLP-3 RT, retatrutide) acts on three receptors: GIP, GLP-1 and glucagon
  • GLP-1 SM (GLP-1 SM, semaglutide) is a once-weekly GLP-1 receptor agonist with a fatty-acid side chain
  • Cagrilintide is a long-acting amylin analog, a fullness signal that runs apart from GLP-1

Metabolic research comes at weight from several directions: incretin receptors, amylin, glucagon, a growth hormone fragment and the mitochondria. Every one of them is in stock in our weight loss peptides.

Checking your vials

Each vial carries the black and white Onyx Research label with the compound, the amount, 99% purity and the research-use line. Check that the name and amount match your order and that the cap and crimp are tight.

Freeze-dried powder often shakes loose in transit. Loose powder is no problem if it’s dry. Droplets on the glass or a shrunken, glassy film mean moisture got in, and we’ll want to hear about it.

Write the arrival date on the box, and add the mixing date and volume later. That short log answers most questions if a result looks odd.

Reconstitution chart and steps

These figures use the 10mg vial. For a bigger vial, multiply by the size difference.

  • Add 1mL: 10mg/mL (1,000mcg per 0.1mL, or 10 units on a U-100 syringe)
  • Add 2mL: 5mg/mL (500mcg per 0.1mL, or 10 units on a U-100 syringe)
  • Add 3mL: 3.33mg/mL (333.3mcg per 0.1mL, or 10 units on a U-100 syringe)

Bring the vial up to room temperature and clean the stopper with an alcohol pad. Inject the bacteriostatic water against the glass wall, not onto the powder, and swirl slowly until it’s clear. Shaking whips up foam and can damage the chain.

Storage and stability

Put sealed MOTS-c vials in the freezer at -20°C if you won’t use them soon. A few days on a shelf during transit does no harm to dry powder. Once you’ve added water, the vial lives in the fridge at 2 to 8°C for up to about four weeks.

Mix one vial at a time. Dry powder keeps for months, but solution only keeps for weeks.
