CJC-1295 DAC background

CJC-1295 with DAC is modified GRF(1-29), the first 29 amino acids of growth hormone releasing hormone with four substitutions, plus a lysine carrying a maleimide linker. That linker is the drug affinity complex, made by ConjuChem in Montreal.

Once in the blood, the DAC bonds to albumin. That takes the half-life from minutes to about a week, which is the whole difference between this peptide and CJC-1295 no DAC.

How CJC-1295 DAC works

The peptide part works like GHRH. It binds the GHRH receptor on pituitary somatotroph cells and switches on the cAMP pathway that drives GH release and GH gene expression.

The DAC linker reacts with a free cysteine on albumin, so the peptide rides around on a protein that lasts weeks. That gives a steady, long GHRH signal instead of the short burst you get from sermorelin or CJC-1295 no DAC.

Published studies

Teichman and colleagues reported in the Journal of Clinical Endocrinology and Metabolism in 2006 that single injections in healthy adults raised GH for about six days and IGF-1 for up to two weeks.

A follow-up paper by Ionescu and Frohman found that GH stayed pulsatile on CJC-1295, with higher baseline levels between pulses. Development stopped after phase 2.

Reconstitution chart and steps

Concentration is simple division: the 5mg in the vial over the volume of water you add. Here’s the 5mg vial at three common volumes:

Use bacteriostatic water, added slowly down the inside of the vial. Swirl, don’t shake. If some powder sticks to the glass, rest the vial in the fridge for a few minutes and swirl again.

Keeping the vial stable

Put sealed CJC-1295 DAC vials in the freezer at -20°C if you won’t use them soon. A few days on a shelf during transit does no harm to dry powder. Once you’ve added water, the vial lives in the fridge at 2 to 8°C for up to about four weeks.

Mix one vial at a time. Dry powder keeps for months, but solution only keeps for weeks.
