IGF-1 LR3 background

IGF-1 LR3 is an 83-amino-acid analog of IGF-1 with an arginine swap at position 3 and a 13-amino-acid extension. Those changes stop it binding IGF binding proteins, so it stays active much longer.

It’s widely used in cell culture media.

What it does at the cell level

IGF-1 is the hormone that carries out much of growth hormone’s effect on cells. Most of it in the body is bound by IGF binding proteins, which hold it inactive.

LR3 has an arginine in place of glutamate at position 3 plus a 13-residue extension. Binding proteins barely recognize it, so it stays free and active far longer. That’s why it’s the standard IGF-1 in cell culture media.

How IGF-1 LR3 compares

Checking your vials

Mixing IGF-1 LR3: a reconstitution chart

Concentration is simple division: the 1mg in the vial over the volume of water you add. Here’s the 1mg vial at three common volumes:

• Add 1mL: 1mg/mL (100mcg per 0.1mL, or 10 units on a U-100 syringe)
• Add 2mL: 0.5mg/mL (50mcg per 0.1mL, or 10 units on a U-100 syringe)

Use bacteriostatic water, added slowly down the inside of the vial. Swirl, don’t shake. If some powder sticks to the glass, rest the vial in the fridge for a few minutes and swirl again.

Storage and stability

Sealed IGF-1 LR3 lasts longest frozen at -20°C and dark. The fridge is fine for short spells. Mixed solution stays at 2 to 8°C and should be used within roughly 28 days.

Let a frozen vial come up to room temperature before you open it, or condensation forms on the powder.
